ABSTRACT

Juvenile zebrafish were used to screen the active components of Lycii Fructus for improving osteoporosis. The screening results were further verified by zebrafish adult osteoporosis model and the action mechanism was explored. Prednisolone was used as the inducer to build osteoporosis models of juvenile and adult zebrafish, and 9 groups of samples of different extracts and chemical parts of Lycii Fructus were given. Alizarin red staining was applied for observing the scale matrix mineralization and bone resorption. The activities of osteoblasts and osteoclasts were detected using alkaline phosphatase (ALP) and tartrate resistant acid phosphatase (TRAP/TRACP) staining. The expressions of bone metabolism-related genes alp, osteoprotectin (opn), osteoblast specific transcription factor (sp7), cathepsin K (ctsk), tracp, and Runt family transcription factor 2b (runx2b) in each group were determined using quantitative polymerase chain reaction. The results showed that all components of Lycii Fructus improved the formation area of the first vertebrae, the staining light density value, and the number of vertebrae joints in juvenile zebrafish and the Lycium barbarum polysaccharide (LBP) treatment group exerted the best effect. In addition, LBP prevented the formation of bone resorption lacunae in zebrafish scales, increased ALP activity, decreased TRAP activity, up-regulated the alp, sp7, and opn genes, and lowered the expressions of ctsk and tracp genes. In conclusion, LBP regulated the activity of osteoblasts and osteoclasts, reduced bone resorption, promoted bone formation and enhanced bone density, which might be the main anti-osteoporosis active fraction of Lycii Fructus. This study provided modern scientific evidence for the scientific connotation of the traditional effect of "strengthening bones and muscles" of Lycii Fructus, provided the reference for the evaluation of the anti-osteoporosis activity of traditional Chinese medicine based on zebrafish adult model, and provided beneficial enlightenment for the bone health needs of the aging society population.

ABSTRACT

ObjectiveTo reveal the effects of different microbial agents on quality of Lycii Fructus by comparing the differences in the contents of multiple types of chemical components in Lycii Fructus after the application of different microbial agents. MethodTaking Ningqi No. 7 as experimental material, four microbial agents, namely Peiyuan combined with Xinterui(TP group), Trichoderma harzianum combined with Bacillus subtilis(BW group), Genwuyou(MT group) and Junyiduo(JYD group), were applied, and no microbial agents was used as the blank group(CK group). Then the contents of total phenolics, total flavonoids, saccharides, amino acids, nucleosides and bases, betaine and other components in Lycii Fructus were determined by ultraviolet spectrophotometry(UV), high performance liquid chromatography(HPLC) and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS/MS), and the methods such as multiple comparisons, principal component analysis(PCA) and partial least squares-discriminant analysis(PLS-DA) were used to analyze the effect of different microbial agents on the quality of Lycii Fructus. ResultMicrobial agents had different effects on chemical components of Lycii Fructus. The content of total phenolics was the highest in the TP group, and it varied significantly from the CK group(P<0.05). The total flavonoid content was the highest in the BW group, followed by the TP group. Both polysaccharide and alduronic acid contents were the highest in the JYD group. Betaine content in the TP and BW groups were significantly higher than that in the CK group(P<0.05). For the determined 23 kinds of amino acids, most of them were the lowest in the JYD group, and the highest in the MT group, while the nucleoside bases were higher in the MT and BW groups. It indicated that Lycii Fructus from different treatment groups could be distinguished clearly based on the determined 45 chemical components. The result of PLS-DA showed that the major differential components in each group were polysaccharides, glucose, fructose, betaine, alduronic acid, asparagine, sucrose, threonine, total flavonoids, alanine and total phenolics. The results of PCA composite scores based on the main differential components showed that composite scores of chemical components in each group were BW group>TP group>MT group>CK group>JYD group. ConclusionThe application of microbial agents of BW, TP and MT can promote the quality improvement of Lycii Fructus, and the application of JYD can promote the accumulation of polysaccharides and alduronic acid to a certain extent, but the overall effect on the quality of Lycii Fructus is not clear. This study lays the foundation for the green and healthy development of Lycii Fructus industry.

ABSTRACT

The chemical components of Lycii Fructus were analyzed by liquid chromatography( LC) and mass spectrometry( MS for the establishment of spectrum-activity relationship,on the basis of which its antioxidant active ingredients were determined. In this experiment,Lycii Fructus was extracted with different solvents and then separated into 80 samples by macroporous adsorption resin and reversed-phase chromatography,respectively. The antioxidant components were enriched into 11 samples and their scavenging abilities against DPPH free radical and ferric ion reducing antioxidant power( FRAP) were significantly stronger than those before the treatment( P<0. 05). The spectrum-activity relationship regarding the antioxidant activity in vitro of Lycii Fructus was established by Pearson correlation analysis,orthogonal partial least squares( OPLS) and elastic net regression. Six chromatographic peaks greatly contributing to the antioxidant activity in vitro of Lycii Fructus were identified as rutin( P6),quercetin( P35),scopoletin( P14),N-cis-feruloyl-4-O-β-D-glucopyranosyl-tyramine or N-( 4-O-β-D-glucopyranosyl-trans-feruloyl)-tyramine( P8), ferulic acid( P13) and1,3,5-dihydroxy-2-isoprenyl-3-xanthone( P23). The active components associated with free radical scavenging were rutin and quercetin both belonging to flavonoids. The reduction of Fe3+was based on phenylpropanoids such as ferulic acid,scopoletin,xanthone and phenolic amides. These results indicated that the antioxidant activity of Lycii Fructus was ascribed to the synergistic action of different products through different ways. Besides,the data analysis model should be chosen carefully for the establishment of spectrum-activity relationship,thus ensuring the reliability of results.

ABSTRACT

Trading volume in Chinese medicinal materials market has gradually declined, and origin market trade has become a trend. Studying the degree of market integration is helpful to understand the operation of origin market of Chinese medicinal materials, which is of great significance for rational planning of the production of Chinese medicinal materials and improving market efficiency. In this study, four varieties (Lonicerae Japonicae Flos, Lycii Fructus, Isatidis Radix and Pseudostellariae Radix) were selected to study the market integration degree of Chinese medicinal materials. Based on the price index data from 2013 to 2019, this paper makes an empirical analysis by using co-integration test, error correction model and Granger causality test. The results showed that the long-term integration of Chinese medicinal materials market was relatively high, but the short-term integration was far lower than the long-term integration. Finally, the author puts forward four policy suggestions: improving the price information platform conduction of Chinese medicinal materials, building high-quality brand of Chinese medicinal materials in non-authentic producing areas, speeding up the establishment quality traceability system and building modern logistics system of Chinese medicinal materials.

ABSTRACT

OBJECTIVE:To investi gate the potential mechanism of couplet medicine of Cuscutae Semen-Lycii Fructus in the treatment of premature ovarian failure. METHODS :Main active components and related targets of couplet medicine of Cuscutae Semen-Lycii Fructus in the treatment of premature ovarian failure were obtained from TCMSP ,GeneCards and OMIM database. The intersection genes between them were screened using Venn online tool. Cytoscape 3.7.0 software was adopted to establish the active ingredients-target network and the PPI network. GO and KEGG pathway enrichment analysis on intersection genes were carried out by DAVID database. Finally ,an active component-target-key pathway network was constructed. RESULTS :Totally 42 active components ,231 and 1 913 targets for active components and disease were obtained from couplet medicine of Cuscutae Semen-Lycii Fructus. The components with high node degree included quercetin ,kaempferol,β-sitosterol,isorhamnetin,glycitein, stigmasterol and sesamin ,etc. There were 149 intersection genes between the active component targets and premature ovarian failure targets. PPI network contained 149 nodes and 2 970 edges,with an average node degree of 39.9 and an average medium of 0.005 4. The results of GO analysis showed that molecule function of the above-mentioned genes mainly involved protein binding , enzyme binding ,etc. Biological process mainly included that positive regulatio n of transcription from RNA polymerase Ⅱ promoter,positive regulation of transcription DNA-templated , Cell components mainly included nucleus ,cytoplasm,etc. Signaling pathway mainly involved cancer signaling pathway , hepatitis B signling pathway ,PI3K/AKT signaling pathway , MAPK signaling pathway , etc. The results of active 617693370@qq.com component-target-key pathway network showed that active components of Cuscutae Semen and Lycii Fructus were flavonoids and alcohols ;key target included AKT 1,TP53, VEGFA,IL6,TNF,etc. Signaling pathway mainly involved cancer signaling pathway ,hepatitis B signaling pathway ,PI3K/AKT signaling pathway ,MAPK signaling pathway ,etc. CONCLUSIONS :Through PI 3K/AKT signaling pathway and MAPK signaling pathway,the active components of couplet medicine of Cuscutae Semen-Lycii Fructus may act on AKT 1,TP53 and other targets , and then play a therapeutic role on premature ovarian failure. The Potential active components stigmasterol ,sesamin and potential targets IL 6,TNF were found.

ABSTRACT

Objective:The chemical differences of Lycii Fructus samples from Qinghai, Ningxia, Gansu, Xinjiang and Inner Mongolia provinces were compared based on proton nuclear magnetic resonance (1H-NMR) plant metabolomics. Method:A total of 97 Lycii Fructus samples from five provinces were collected, including 61 samples in Qinghai, and extracted by 50% methanol for detecting. 1H-NMR spectra were obtained and compared by multivariate statistical analysis for investigating the chemical differences of samples from Qinghai and other production areas. And the content of Lycii Fructus polysaccharides in all samples was determined with the wavelength of 490 nm (calculated by anhydrous glucose). Result:A total of 32 chemical components were detected in the Lycii Fructus extract by 1H-NMR. The multivariate statistical analysis revealed that there was no significant difference among the samples from five provinces. The difference between Lycii Fructus from Qinghai and Ningxia, as well as the samples among the six regions of Qinghai province were relatively small. The similarity values of the majority of samples were >0.85. Univariate analysis showed that no significant difference was observed for the most metabolites in Lycii Fructus collected from five provinces, except for sucrose, glucose, proline and so on. There was no significant difference in the content of Lycii Fructus polysaccharides between Qinghai and other provinces. And the correlation coefficient between the content of Lycii Fructus polysaccharides and the small molecular compounds identified by 1H-NMR was -0.2-0.4. Conclusion:In this study, chemical characteristics of Lycii Fructus in Qinghai province are analyzed from the holistic view by 1H-NMR plant metabolomics, in combination of polysaccharide determination, and the results show that there is no significant difference between samples from Qinghai and other four provinces. The quality evaluation method based on 1H-NMR established in this study can provide scientific basis for improving quality control level and selecting planting areas of Lycii Fructus.

ABSTRACT

Objective:To explore the molecular mechanism of Lycii Fructus and Salviae Miltiorrhizae Radix et Rhizoma (FLRSM) in the treatment of retinitis pigmentosa (RP) based on network pharmacology and bioinformatics. Method:Possible intake active components and targets of FLRSM were screened out and predicted by traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP). Gene targets related to RP were mined through disease gene databases. Protein-protein interaction (PPI) network of component-targets and disease-targets were mapped by functional protein association networks (STRING), and the intersection of the two networks was obtained. The gene ontology and kyoto encyclopedia of genes and genomes(KEGG) pathway of the intersection network were analyzed by the database for annotation, visualization and integrated discovery(DAVID). CytoHubba analysis was used to screen out the key targets. Result:A total of 390 active ingredients related to FLRSM were retrieved from TCMSP. According to pharmacokinetic parameters, 110 active ingredients were screened out, 19 active ingredients were further screened out, and 208 targets related to these constituents were retrieved. Totally 206 genes directly related to RP were obtained from the disease gene databases. And 79 genes were obtained from the intersection of PPI networks of component targets and disease targets. These genes mainly involved in biological processes, such as protein autophosphorylation, transcriptional regulation and cell proliferation, and the molecular functions mainly involved adenosine triphosphate binding, transcription factor activity, core promoter binding, and were enriched in nuclear, transcription factor complex, nucleus, cytoplasm and other regions. It was mainly related to neurotrophin signaling pathway, cell cycle related pathway and Wnt signaling pathway. And 8 key gene targets for FLRSM treatment of RP were identified by further screening. Conclusion:The material basis of pharmacodynamic action of FLRSM involves 19 active ingredients, such as porous sterol and tanshinone ⅡA. The key targets of FLRSM in the treatment of RP include 8 genes, such as E2F transcription factor 1(E2F1) and retinoblastoma gene1(RB1).The main mechanism is related to the regulation of neurotrophin signaling pathways, cell cycle related pathways and other signaling networks.

ABSTRACT

Objective: To investigate the inhibitory effect and mechanism of the extracts from fresh Lycii Fructus (LBL) on hepatoma HepG2 cell-induced cachexia in mouse.Method: The human hepatoma cell line HepG2 was injected into BALB/C mice to establish the cachexia model.Then the LBL was fed to the models respectively in low dose (5 mg·kg-1) or high dose (25 mg·kg-1).After 28 days of continuous feeding,the mice's body weight was detected.The expression levels of creatine kinase (CK),interleukin-1β (IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were evaluated by enzyme linked immunosorbent assay (ELISA).The muscle degradation and the expression of p38 mitogen-activated protein kinase (p38 MAPK) were detected by immunohistochemistry staining.The expression levels of phospho-p38 mitogen-activated protein kinase (p-p38 MAPK) and nuclear factor-κB (NF-κB) were determined by Western blot.Result: Compared with cachexia group,the loss of body weight and muscle decomposition were significantly inhibited both in the low dose LBL group and high dose LBL group (P0.05,PPβ,IL-6 and TNF-α in both the low-dose LBL group and the high-dose LBL group (PPPκB were inhibited both in the low-dose LBL group and the high-dose LBL group (PConclusion: LBL could inhibit the cachexia induced by hepatoma HepG2 cell line in mouse,suggesting LBL could reduce major cytokine and plasma inflammatory factors through p-p38 MAPK pathway.

ABSTRACT

Objective:To prepare Lycii Fructus polysaccharide buccal tablets and investigate its immunomodulatory effect. Method:Taking the appearance, taste, hardness and disintegration time of the tablets as comprehensive evaluation index, based on single factor tests, central composite design-response surface methodology was adopted to optimize the prescription of Lycii Fructus polysaccharide buccal tablets with mass ratio of dextrin to mannitol, mass ratio of cyclamate to malic acid and dosage of sodium carboxymethyl starch (CMS-Na) as factors. Kunming mice were randomly divided into 5 groups, namely the Lycii Fructus polysaccharide buccal tablets low (100 mg·kg-1·d-1), medium (200 mg·kg-1·d-1) and high (300 mg·kg-1·d-1) dose groups, the normal group (0.9% normal saline, 300 mg·kg-1·d-1) and the positive medicine group (Cinengsu group, 300 mg·kg-1·d-1). The immunomodulatory effect of the buccal tablets were investigated by calculating immune organ index, monocyte-macrophage phagocytic index, serum hemolysin antibody level, and the voix pedis thickness difference of delayed hypersensitivity (DTH) of mice. Result:Optimal prescription for the buccal tablets was 80% of Lycii Fructus extract, 11.5% of dextrin-mannitol (1.2:1), 1% of cyclamate-malic acid (1:1), 0.5% of cream essence, 6.5% of CMS-Na, 0.5% of magnesium stearate, and appropriate amount of 80% ethanol. Under the optimal condition, the hardness of the buccal tablets was 11.83 kg, its disintegration time was 13.21 min, both of which were in line with the relevant provisions of the 2015 edition of Chinese Pharmacopoeia, and the buccal tablets had good appearance and taste. Compared with the normal group, medium and high dose groups of Lycii Fructus polysaccharide buccal tablets significantly increased thymus index, spleen index and phagocytic index of mice (PPPPPConclusion:The formulation process of the buccal tablets optimized by central composite design-response surface methodology is stable and feasible, and Lycii Fructus polysaccharide buccal tablets can improve the immune regulation function of normal mice, and this study can provide experimental basis for the development, utilization and clinical application of Lycii Fructus and Lycii Fructus polysaccharides.

ABSTRACT

Objective: To optimize the extraction process of effective components in Lycii Fructus using central composite design(CCD)-response surface method(RSM). Methods: The yield of main effective components, polysaccharides, flavonoids, rutin and chlorogenic acid as well as the yield of total extract were used as indicator for evaluating the effect of extraction solvent, extraction time, solid- liquid ratio, extraction temperature and other factors. The single factor experiment was conducted to determine the range of various factor parameters, the factor design experiment was performed for the screening of the factors that had significant influence on the extraction efficiency of effective components, the maximum velocity rise experiment was carried out to determine the central value of the factors, and finally the optimal extraction process was optimized by the central composite design- response surface methodology. Results: The optimum extraction conditions of Lycii Fructus were obtained as follows: the extraction with 60% aqueous ethanol solution by ultrasound sonication at room temperature for 40.5 minutes, and the solid- liquid ratio was 1:27. Conclusion: The present optimal extraction process could effectively improve the extraction efficacy of effective components from Lycii Fructus, and the created CCD-RSM model and related conditions have good predictability and repeatability, which provides experimental and theoretical reference for further development and research of Lycii Fructus.

ABSTRACT

Lycii Fructus (Lycium barbarum, Gouqizi in Chinese name) is one of the most popular Chinese material medica and a common ingredient in tonic food. This fruit has been paid rapidly growing attention for its nutrient value and noticeable pharmacological properties. The present paper focuses on the legal resource of Gouqizi and reviews the main research in medicinal field including botanical identification, ethnopharmacological functions, phytochemistry, pharmacological effects, clinic usages, and safety issues. In addition, some issues needed address will be also discussed. We strongly believe that further investigation will deepen our knowledge of Gouqizi and promote the industrial development in the world.

ABSTRACT

Objective To mitigate Lycii Fructus surface crust and save drying time during drying process, vacuum pulsed drying technology was applied to dry Lycii Fructus so as to investigate moisture diffusion regulation and build the drying model. Methods The effect of different drying temperature (50, 55, 60, and 65 ℃), vacuum pressure holding time (5, 10, 20, and 30 min), and atmosphere pressure holding time (2, 4, and 8 min) on drying characteristics, moisture diffusion coefficients and drying activation energy was explored in vacuum pulsed drying process. Weibull model distribution was used to simulate and analyze Lycii Fructus drying curves. Results All the drying temperature, atmosphere pressure holding time, and vacuum pressure time holding time had significant influence on drying time. When drying temperature, atmosphere pressure holding time, and vacuum pressure time holding time was 60 ℃, 4 min, and 10 min, respectively, the minimum drying time was achieved to be 284 min. Weibull distribution model can be well described the vacuum pulsed drying process of Lycii Fructus. The scale parameter was related to drying time and decreased as drying temperature increased. The drying temperature, atmosphere pressure holding time, and vacuum pressure time holding time had little influence on the shape parameter. The shape parameter was associated with drying method. The moisture diffusion coefficient and activation energy were calculated to be 2.02 × 10-8-3.56 × 10-8 m2/s and 36.27 kJ/mol, respectively. Conclusion Weibull distribution model can well describe the moisture diffusion regulation of vacuum pulsed drying process of Lycii Fructus. The drying result had a great significance for predicting, controlling and optimizing drying process. On the other hand, the research could provide technical basis for industrial drying of Lycii Fructus by vacuum pulsed drying technology.

ABSTRACT

Objective Three kinds of pretreatment methods were studied to compare and select the suitable pretreatment methods for Lycii Fructus (LF) reference standards to improve its crushing and packaging efficiency. Methods LF was treated by three pretreatment methods of decompression drying, liquid nitrogen freeze-drying, and freeze-drying. The difference of characteristic values before and after treatment was determined and compared with LF without pretreatment. The determination of characteristic values included the determination of moisture, content, etc. Results The water content of raw material of directly crushed LF was 6.4%, and the water content of the other three pretreated LF was 4.6%, 5.0%, and 5.2%, respectively. The content of polysaccharide and betaine in the raw material of directly crushed LF was 2.5% and 0.98%. The polysaccharide content of the other three pretreated LF was 2.6%, 2.8%, and 2.6%, respectively. The content of betaine was 0.94%, 1.00%, and 0.87% by the other three pretreatment methods. The results showed that there was no significant difference in the quality of LF obtained by the three pretreatment methods. Therefore, from an economic point of view, this paper recommended to use more affordable pretreatment method of decompression drying for LF. In addition, the pretreated raw materials of LF should be crushed and subpackaged as soon as possible, and the crushing and the dividing environment humidity should be strictly controlled at 20% to 30%, in order to improve the efficiency of crushing and dividing. Conclusion Based on the cost consideration, the decompression drying method is the most economical and effective of the three pretreatment methods. Therefore, the decompression drying method is recommended in actual work.

Objective To optimize the extraction technology of total flavonoids from Lycii Fructus by response surface methodology (RSM).Methods On the basis of single-factor tests, enzyme concentration, enzymolysis time, and enzyme solution temperature were selected as influencing factors during extraction to conduct three-factor three-level center combination design. The effects of three factors on the yield of total flavonoids from Lycii Fructus were analyzed by RSM.Results The optimum conditions for extraction technology of total flavonoids from Lycii Fructus were enzyme concentration 0.30%, enzymolysis time 1.0 h, enzyme solution temperature 60℃. The predicted extraction yield of total flavonoids was 0.9492%.ConclusionUsing RSM to optimize the extraction technology of total flavonoids from Lycii Fructus is stable, feasible, and simple, which can provide references for further study and application.

In this review, the new technology application in extraction, purification, and structural elucidation of Lycii Fructus polysaccharide was systematically untangled, the current research situation and development trend were outlined, and the potential product groups and industry prospect in Lycii Fructus polysaccharide industrialization were analyzed to provide scientific reference for the enhancement of effective utilization and industrial chain of Lycii Fructus resources.

AIM To determine the contents of chemical consituents in Lycii Fructus from Qaidam Basin.METHODS Spectrophotometry was adopted in the content determination of polysaccharides,total flavonoids and carotenoid.HPLC was applied to the content determination of betaine and scopoletin.Kjeldahl method was used for the content determination of protein.Then principal component analysis was performed.RESULTS The contents of carotenoid,betaine and scopoletin in samples from six growing areas showed obvious differences (P < 0.05),while those of polysaccharides,total flavonoids and protein exhibited no obvious differences (P > 0.05).The contents of various constituents in samples at three picking time also had no obvious differences (P > 0.05).The comprehensive score of principal components of samples from Delingha City was the highest,followed by that from Ulan County.CONCLUSION The quality of Lycii Fructus from Qaidam Basin from Delingha City is the best.

ABSTRACT

Lycii Fructus is a traditional medicinal and edible herb, with the function of liver and kidney nourishing, blood and eyesight replenishing. As the most important active substance in the fruits of Lycium barbarum, Lycium barbarum polysaccharides (LBP) have been demonstrated to play multiple pharmacological activities, with broad prospects for development and utilization. Based on the comprehensive deep analysis of global LBP patent output, the current patent LBP features were explored from the perspective of development trend, technology field distribution, time dimension, technology life curve and patent applicant. The development trend of Chinese LBP industry was also revealed. At present, the research and development of LBP is in the "development period", with a good development track in which the main research institutions remain domestic ones. At the same time, problem still presents in the lack of industrialization, which means that the advantage of natural resources has not been transformed into industrial advantages of LBP. The format of potential product group and prospect of LBP were also analyzed to provide scientific information for the effective development,comprehensive utilization and collaborative innovation mechanism of Chinese Lycium barbarum resources and LBP.

ABSTRACT

The distribution information of Lycii Fructus was collected by interview investigation and field survey, and 46 related environmental factors were collected, some kinds of functional chemical constituents the of Lycii Fructus were analyzed. Integrated climate, topography and other related ecological factors, the habitat suitability study was conducted based on Arc geographic information system(ArcGIS),and maximum entropy model. The AUC of ROC curve was both above 0.95, indicating that the predictive results with the maximum model were highly precise. The results showed that 5 major ecological factors had obvious influence on ecology suitability distributions of Lycii Fructus, including soil pH, soil subclass, vegetation type and in August the average temperature et al. It is suitable for the living habits of the Lycii Fructus, dry, cool weather, more hardy, drought-resistant, alkali soil, which is suitable for distribution in the northern temperate plains. In addition, the ecological suitability regionalization based on the chemical constituents of Lycii Fructus also provides a new suitable distribution area other than the traditional distribution area, which provides a scientific basis for the reasonable introduction of Lycii Fructus.

ABSTRACT

OBJECTIVE: To establish an HPCE method for determination of N-trans-feruloyl tyramine in Lycii Fructus. METHODS: The chromatographic separation was performed on an uncoated fused silica capillary of 50 cm × 75 μm ID (40 cm of effective length). 50 mmol · L-1Na2B4O7(pH 9.0 with H3PO4) was selected as the running buffer. The applied voltage was 20 kV, and the column temperature was maintained at 25°C. The chromatogram was detected at 214 nm. RESULTS: The linearity for N-trans-feruloyl tyramine was in a range of 0.25 - 15 μg · mL-1 (r = 0.9998, n = 7). The average recovery for N-trans-feruloyl tyramine was 101.8% with RSD of 2.06%. CONCLUSION: The method is accurate, reliable, and can be used as a quality control method for N-trans-feruloyl tyramine in Lycii Fructus, which can provide basis for development and utilization of Lycii Fructus. ABSTRACT

The principal objective of this study was to evaluate the quality characteristics of mungbeanmook prepared with five different levels (0, 5, 10, 15, and 20%) of Gugija (Lycii fructus) infusion. We analyzed Hunter's color values, pH, texture characteristics, and perfomed sensory evaluation of mungbeanmook prepared with Gugija infusion. The pH levels of mungbeanmook ranged between 5.45 and 6.12 and did not show significant differences. As Gugija infusion concentration increased, the color lightness "L" value decreased, whereas redness color "a" value and yellowness color "b" values increased (P<0.05). With regard to mechanical texture properties of the mungbeanmook samples, as the Gugija infusion concentration increased, the scores of hardness, chewiness, and gumminess all significantly decreased (P<0.05) whereas there were no significant differences in springiness or cohesiveness. The results of the sensory evaluation showed that the score of the LFLT10 sample (10% Gugija infusion) was significantly higher than those of others in terms of taste, color, flavor, texture, and overall quality (P<0.05). Taken together, the recommended optimum level of Gugija infusion in mungbeanmook is 10% for sensory evaluation. Gugija infusion can be a useful approach to improving quality of mungbeenmook as a functional food.
